Experience:

Intoduction to culture media and types

Culture media:


Most laboratoris purchase their media in a dehydrated form or reday to use.
Many culture media have generic name and and should not be assumed to be identical as the individual manufacture ingredients have unique characteistics.
Some culture media have proprietary names and is available only from particular companies.
Though can be purchased from others 'equivalent " items should not be considered to be identical.

Only two commercial available media are rigorously controlled to meet external regulatory specifications.
Standard Plate Agar and mueller-hinton agar.

General purpose media
These are non selective media that are designed to supportthe growth of ide variety of heterotrophic bacteria
This media contains ezyme or acid digests of animal or plant proteins as prime sources of organic carbon and nitrogen
Metal ions, vitamins, minerals re provided by yeast extracts / meat extracts.
The groth of more fastidious organisims is encouraged by growth promoting supplements.
Growth promoting supplements include horse/ sheep blood, Fides extract, Menadione/vitK, Ferric/Ferrous salts, vitamins, Horse/Rabbit serum, Yeast Autolysate, Sodium Pyruvate.

Selective isolation media.
This media is used for the isolation of certain micro-organisms and contains selective chemicals that supress the growth of others.

Usually the suppresed groth is during the early hrs of incubation which allos the slower variety to grow which are the unsuspected organisms.

Transport media
Semisolid media with/ without charcoal and antibiotics intended to keep delicate microbes alive during transit.

Antibiotic sensitivity media
Mueller-Hinton is the most widely used AST media. Isosensitivity and Direct Sensitivity test agars are used fro disc diffusion test or antibody dilution.

Sterility test
For products containing presevatives: Cluasen medium, Thioglycollate media.
For products not containing presevatives: Any non- selective broth medium.

Introduction to sterilization , disinfection.

Sterilization and Disinfection.


 Sterilization means complete destruction of all microorganisms including spores.

 

Disinfection means destruction of vegetative organism that might cause spoilage or diseases.

 It neccessarily dosent mean killing of spores.

 
Decontamination
Decontamination is the process of cleansing to remove contamination, or the possibility (or fear) of contamination.
 
 Sterilization can be caused by many techniques
the methods are :
  1. red heat
  2. dry heat
  3. steam under pressure(autoclaving)
  4. steam not under pressure(tyndallization)
  5. filtration
  6. inceneration
disinfection
chemical disinfection:
  1. phenolics
  2. hypochlorites
  3. alcohol
  4. formaldehyde
  5. gluteraldehyde
  6. iodophores
  7. QAC

Types and uses

 The toxicity, harmfull effects and spectrum of suceptibility should be considered while using the disinfectants.

 

 Testing of disinfectants

 Several countries and organisations have offixcial tests for disfectants here the specifications are detailed.

 The basic test involves having a suspension of known colony forming units (cfu/ ml) of Staphylococcus aureus/Psuedomonas aeroginosa and then testing it with several known dilutions of of the disinfectant to be tested.

 After a pre determined contact time aknown volume is removed and neutralizing agent is added that deactivates the disfectant in question.

 The surviving colony forming unite are then calculated.

 A good disfectant reduces the colony forming units by 5 log units in an hour.

 

Usually the best test is usually left to the manufacturers but one test thats simple and easy to use is "in use test".

    2.   Molecular and Cellular biology:

     3.  Chemical And Synthetic Biology:

     4.  Zebra Fish Biology:

          Applicant (s) should be well versed with modern molecular biology and have working experience on chemical genomics using zebra fish as a model system.


     5.  Structural Biology:
         applicants background: Applicant(s) should have a demonstrated record of significant accomplishment in the areas of X-ray crystallography /NMR spectroscopy/ EM and optical imaging.


         

     6.   Skin Biology:
          applicants background: Applicant(s) should have interest in understanding the tissue level dynamics of     skin and expertise in using diverse approaches to study mechanisms of skin diseases / biology.


         
     7.   Asthma Immunology:
           applicants background: Applicant(s) should have research interests in molecular and cellular basis of asthma especially in the area of epithelial biology such as airway remodeling.


          

     8.   Tuberculosis research:
            applicants background: Applicant(s) should have extensive experience in studying host-pathogen interactions in the context of tuberculosis with a commitment to manage BSL-3 facility and specialized animal facility at IGIB.
